DNA-TA-21_NV - a family of really unusual transposons from the starlet sea anemone genome.

Abstract:
Originally, this transposable element was reported under name REP5, composed of the REP5-xLTR_NV and REP5-xI_NV parts [1]. Later on, we renamed these to DNA-TA-21_NV, DNA-TA-21-LTR_NV and DNA-TA-21-I_NV, respectively. The genome contains numerous solo copies of DNA-TA-21-LTR_NV, inserted into CTAG target sites (accompanied by the TA target site duplications). In addition to the solo LTRs, the genome contains many copies of complete DNA-TA-21_NV elements, composed of the internal portion flanked by its two LTRs. Again, the complete elements are also inserted into the CTAG target sites, with the TA TSDs. Finally, the genome contains also structures like LTR-I-LTR-I-LTR. On the one hand, similar structures are typical for LTR retrotransposons. However, DNA-TA-21-I_NV does not have a 3' polypurine tract and 5' primer binding site that are typically present in autonomous and non-autonomous LTR retrotransposons. Moreover, DNA-TA-21-LTR_NV contains 33/32-bp TIRs, analogous to DNA transposons. Also, known LTR retrotransposons do not produce 2-bp target site duplications. Therefore, we classify this transposable element as a DNA transposon. We predict that DNA-TA-21_NV belongs to a novel group of transposons mobilized by autonomous elements with uknown enzymatic activities.
