Abstract:
HELITRON1_AG is a first example of HELITRONs (a rolling-circle DNA transposons) present in insects. The A. gambiae genome contains many families of HELITRONs. HELITRON1_AF is one of them. A defective copy of a HELITRON-like element is also present in the D. melanogaster genome (AE002840, positions 9827-9264). HELITRON1_AG has all basic hallmarks of HELITRONs: it is flanked by 5'-TC and CTAG-3', is inserted into the AT target site and encodes a 1792-aa HELITRON-like Rep/Helicase protein, called AGHEL1p. AGHEL1p (positions 820-6195): MQPSVSDIVSQNLTPPEETPNEKKARLQRKRQALYRAKKRLPGAAPTLAAVQDDQQAVPSTLAGSSLSAA SAILQQQQRRNTDDELLNEIAGRYSTPPDETPPEKKARLARKRQALYRAKKRLAGAAPTVAAVQDDQQAV PSTSAGTSLSGAPAILQQQQLRRNIDDDLLNEIAGRYSTPPDETPPEKKARLARKRQALYNARKRMVQTP NVATVVNRAPAPASAPVPAPAPAPTPALVDPTAAVAIRVPIHQQLQTYVRHLRDHEADQQRQFIARRRMT GHLRVAHGNHESYSCPSLHTLAPRVPCNFCSALKWPGEPPSVCCNSGQVVLPPFPEPPAELRQLFDVPQF LLNIRRYNNAFAFTSIGASIRGNDPVRQDLRVGHGGIYNYRIQGALCHRIGSLAVLPGRPPCYAQLYFYD SSSEGQYDEMLNARAKAYGSELNREILAILQRVFSTHNPIAEMFKHAYERMSVRDDLRLCIHSRIPGIDQ RRYNAPTADEVGGVFVCDDTTGATEHTRDIVLQHRATGYLQPVFDTNQLADALQYPILFPRGETGWTYGM PKVQRRTRQQQQPSRPTSSSNGAAHIDENDDAAEDPNPELDGAERSANQITPREYAAYRIAWRENAQTLM HRAGRLFQQYCVDQYCKIEMQRLKYLREHQAQLRTEAYTGFMDLAGAEGTIADLHPDNLPNVAVEIPVVQ EPPVAPRRSIIDPPSNLSRTGTRVILGPSFVGSNRYMRAQYQDAMAIVRALGKPDLFITVTCNPKWPEIT QCLLPRQQAPDRPDVIVRVFRLKLKAILNDLTMGALGIEVARIHVIEFQKRGLPHAHILVILAEEDKPQT PADYDKIVSAELPNPATSSQLFETVQSCMMHGPCGAANPAAPCMKDGTCEKGFPKSFCEQTRSMDNGYPQ YRRRNNGRSVTVKGIELDNRYVVPYNPWFTHKYNCHINVEVCTSISSVKYLYKYVYKGHDRLSVTLAVGN DEIQQHIDARYLSPTDSCWRIMRFELQAKTHTVVTLPIHLENQQNVFFRANETVSCVLNRGNHTMLTRFF QLAAHDNFARALLYHDVPTYYRYAKPTANQRLPWQEPGTKHWIRRIRTGHKTVIGRMVSCSMQLMERYCL RLLLCYRKGPTSYEDLRTVNGSVCETFQQAAINEGLLEDDSEWDRALEEAATYRMPAQLRHFFALILSSG MPQNPRTLWESYASEMSEDFHHRNRDRYTTEESDLNKLLRDVEHFRALSDIDRYLRGTTPSKVLTSSPGM PQLSEYEHVQAHIMDDDDVNEFIIAERSYLITDLDATLATVHQLNDEQRMVYETVTAAIDRQLATAASQA NAGDQRLFFLDGPGGTGKSFLVEKILAHVRRCGEIALATAASGIAALLLTGGKTVHSTFKLPLDLNNHST CSITVQSKRAEMLRQTALIVWDEASMSSRFALEAVDRTLQDITGVQLPFGGKVVLLSGDFRQILPIVPKG TDAQIINECIKKSTLWPLFRSLQLRDNMRVRTAPNANQASELRDFANLLLRIGEGRHDTFAGLDPSLAKI PHDMIVPHTANPTNDLNTLIDKIYPDMQRHFQHPSFFSDRAILSPLNVDVASVNNLVLDRIPGPEQEYRS VDTLVNPEEHEHLQLPSEYLNTLNVSGIPVHRLRLKRFAPVLLLRNLNSDMGLCNGTRLQIVGLKRNCLH AKILTGTRRGEDVLLPRIFCDSNDKGHPFQIRRKQFPVQVCFAMTINKSQGQSLHHLGLYLPQDVFAHGQ LYVALSRVTSRANIAVLIPNPKRADEEGVSTSNIVYREVVDR
